Effects of Hypoxic Preconditioning and Vascular Endothelial Growth Factor on the Survival of Isolated Primary Retinal Ganglion Cells

چکیده
This study aimed to investigate the effect of hypoxic preconditioning (HPC) on primary retinal ganglion cell (RGC) survival and associated mechanism, including role vascular endothelial growth factor (VEGF). Retinas were separated from enucleated eyeballs Sprague–Dawley rats postnatal days 1–4. RGCs harvested using an immunopanning-magnetic separation system maintained for 24 h in a defined medium. Hypoxic damage (0.3% O2) was inflicted cells CO₂ chamber. Anti-VEGF antibody (bevacizumab) administered exposed conditions, RGC rate compared that non-anti-VEGF antibody-treated RGCs. HPC lasting 4 significantly increased rate. In conditions h, VEGF mRNA protein levels increased. Treatment with high dose bevacizumab (>1 mg/mL) countered HPC-mediated survival. Protein kinase B focal adhesion 4-h hypoxia-treated showed beneficial effects However, only specifically controlled exposure rendered neuroprotective effects. Strong inhibition inhibited These results indicate may play essential promoting under conditions.

Effect of Anti-vascular Endothelial Growth Factor Antibody on the Survival of Cultured Retinal Ganglion Cells
PURPOSE To investigate the effects of anti-vascular endothelial growth factor (VEGF) antibody on the survival of retinal ganglion cell (RGC)-5 cells differentiated with staurosporine under oxidative stress. METHODS We used real-time polymerase chain reaction and Western blot to confirm the expression of VEGF, VEGF receptor (VEGFR)-1 and VEGFR-2 in RGC-5 cells differentiated with staurosporine...

PURPOSE To determine the mechanistic role for adenosine and adenosine receptors in the hypoxic induction of vascular endothelial growth factor (VEGF) in retinal microvascular cells. METHODS Bovine retinal capillary endothelial cells and microvascular pericytes were studied under normoxic (95% air, 5% CO2) or hypoxic conditions (0% to 2% O2, 5% CO2, 93% to 95% N2) using a variety of well-chara...
متن کاملBevacizumab neutralizes the protective effect of vascular endothelial growth factor on retinal ganglion cells
PURPOSE Vascular endothelial growth factor (VEGF) is well known for its role in pathologic neovascularization, including wet age-related macular degeneration. However, a growing body of evidence indicates that VEGF is also neuroprotective of non-vascular cells in various animal models through reduction of oxidative stress. In light of the widespread use of intraocular anti-VEGF therapies for ag...
